Для цільових каркасних ядер будуть розроблені препаративні методи синтезу та функціоналізації. Структурні, фізико-хімічні та ADME-tox параметри отриманих похідних буде вивчено за допомогою інструментів комп’ютерної хімії (DFT та ab initio розрахунки) та експериментально. З синтезованих сполук будуть отримані прямі аналоги відомих лікарських засобів на основі адамантану та бензену. Їх біологічна активність буде порівняна у серії тестів, що дозволить однозначно визначити придатність похідних поліциклічних каркасів в якості біоеквівалентів/ біоізостерів адамантану/бензену. На основі всіх даних отриманих в ході дослідження буде створено інтегрований інструментарій для оцінки перспективності та потенційних меж застосування будь-яких нових поліциклічних каркасних сполук для потреб пошуку нових лікарських засобів.The project aims to develop methods for the synthesis and comprehensive study of the properties (structural, physicochemical, ADME-tox, and biological) of polycyclic scaffold compounds. Preparative methods for synthesis and functionalization will be developed for target scaffold nuclei. The structural, physicochemical, and ADME-tox parameters of the obtained derivatives will be studied using computational chemistry tools (DFT and ab initio calculations) and experimentally. Direct analogues of known adamantane- and benzene-based drugs will be obtained from the synthesized compounds. Their biological activity will be compared in a series of tests, which will allow to unambiguously determine the suitability of polycyclic skeleton derivatives as bioequivalents/bioisosteres of adamantane/benzene. Based on all the data obtained during the study, an integrated toolkit will be created to assess the prospects and potential limits of the use of any new polycyclic framework compounds for the search for new drugs.Фундаментальне дослідження
